Intended Use :
Diagnoslic regant for quantitative in vito determination of Creatinine in human serum, plasma and urne by  by Jaffe's method
Clinical Significance :
Creatinine is a waste product formed in muscle from thehigh energy storage compound, creatinine phosphate. Theamount of creatinine produced is fairly constant (unlikeurea) and is primarily a function of muscle mass. 
lt is notgreatly affected by diet, age, sex or exercise.  creatinine isremoved from plasma frcm plasma by glomerular flieraiion and then excoriated in urine without any appreciable resorption by tubules.
Creatinine is used to assess renal function, howeverserum creatinine levels do not start to rise until function has decreased by at least 50%

Methodology :Modified Jaffe's Reaction.
Principle :
Creatinine reacts wilh alkaline picrate to produce anorange-yellow colour ( the Jaffe's reaction ) Specifily ofthe assay has been improved by the introduction of aninitial rate method. However, Cephalosporin antibioticsarc still major interferants.
The absorbance of the orange-yellow colour formed is directly proportional to creatinine concentration and is measured photometrically at 500-520 nm
Reagent PreparationReagent (R1 & R2) are ready lo use. Mix equal volume of Reagent (Ri & R2) lo prepare working reagent.
Storage And Stability :
Unopened Reagent 1,2 and standard arc stable till the expiry date stated on the label.
Keep the standard vial plugged after use in order to avoid deterioration.
Note :
The reagent and sample volumes may be altered propotionally to accommodale vrious analyzer require ments.
The temperature of the kinetic assay must be maintained costant as the rate of colour development is strongly temperature sensitive.
Gross haemolysis causes falsely elevated results.
